John Le Mesurier est né John Elton Halliley le 5 avril 1912 à Chauser Road, Bedford - Angleterre.
Après être parti de l'école, John part travailler pour un cabinet de notaires appelés Greene et Greene, et six ans après, juste avant son 21ème anniversaire, John signa pour le Fay Compton Studio of Dramatic Art, en utilisant le premier nom de sa mère, Le Mesurier, en tant que nom d'artiste.
Quelques mois plus tard, il signe son premier engagement professionnel au Palladium Théatre à Edimbourg.
En 1940, John rejoint le Royal Armoured Corps à Tidworth - Salisbury Plain où il atteint le grade de pilote. Il sert son pays chez lui et en Inde et par conséquent, doit mettre sa carrière de côté jusqu'à se qu'il soit démobilisé en 1945.
En 1946, tout en visitant le Players Theatre à Villiers Street, près de Charing Cross, il rencontre Hattie Jacques.
En 1948, John tourne son premier film, la "Escape From Bradmoor".
John et Hattie se marièrent en avril 1952 et ont eu deux fils, Robin et Kim.
John apparait pour la première fois dans l'émission "Hancock's Half Hour" de la BBC-TV en 1957. Il y apparaitra à de nombreuses reprises.
Après avoir tourné le film "Our Man in Marakesh", John tourna alors le classic TV show « George and the Dragon avec Sid James et Peggy Mount.
La série pour laquelle John est peut-être le plus célèbre est pour le rôle du Sergent Arthur Wilson dans « Dad's Army ». John devait initialement jouer le pilote et Arthur Lowe, le sergent mais les rôles ont vite été inversés.
